胰腺癌生物模型的应用现状与展望

Current State and Future Perspectives of Biological Models in Pancreatic Cancer

摘要: 胰腺癌恶性程度高、早期诊断困难且易转移,患者就诊时常因病灶局部进展而失去根治性手术时机,虽然在综合治疗效果方面已取得明显提高,但患者生存率依然很低,预后差。 胰腺癌的隐匿发病使研究者无法及时获取早期病灶进行深入研究,因此,构建合适的生物模型,对研究胰腺癌发生发展的分子机制并寻找新的治疗策略至关重要。 目前常用于胰腺癌临床前研究的生物模型包括自发诱导模型、移植瘤模型、类器官模型等,本文对各模型的 应用现状进行综述。

Abstract: Attributable to its highly malignant feature, late diagnosis and early metastasis, pancreatic cancer patients often lost the opportunity of radical surgery due to the local advanced lesion. Although significant progress has been made in the effect of comprehensive treatment, the patients were still with poor overall prognosis and low survival rate. The insidious symptoms of pancreatic cancer limited investigators to acquire early tissue timely for in-depth study, therefore, a suitable biological model should be constructed to investigate the molecular mechanisms underlying pancreatic cancer initiation and progression, and to evaluate new therapies. Spontaneous tumor animal models, transplanted tumor models and pancreatic cancer organoid were the most commonly used preclinical biological models, and we reviewed the current trends in applications of biological models in pancreatic cancer in this paper.
